The world record, 46.91, was set at the 2009 World Championships at the height of the now-illegal rubberized supersuit era by Brazil's Cesar Cielo; he did not qualify to race in front of a home crowd in any event at the 2016 Olympics.

Fellow American Caeleb Dressel – whose parents and family made it down to Rio thanks to a secret, surprise GoFundMe campaign set up by the Dressel siblings – finished sixth with 48.02. Dressel was part of the 4x100m freestyle relay that won gold alongside Michael Phelps, Adrian and Ryan Held.
